چکیده

The reversible, mitochondrial membrane-associated transhydrogenase from the midgut of Manduca sexta (L.) (Lepidoptera: Sphingidae) catalyzes hydride-ion transfer between NADP(H) and NAD(H). The effects of ecdysone and 20-hydroxyecdysone were evaluated and compared to both the NADH-NADP and NADPH-NAD transhydrogenations. In the direction of NADPHformation, the developmentally significant transhydrogenations occur as non-energyor energylinked reactions. The energy-linked activity couples with either electron transport-dependent NADH or succinate utilization, or ATP hydrolysis by Mg-dependent ATPase. Upon the addition of ecdysone alone, all energy-linked reactions in the direction of NADPH formation exhibited a notable increase in activity level over the control reaction. The addition of 20hydroxyecdysone yielded no significant increase in the activity of any of the transhydrogenations. Synergistic addition of both ecdysone and 20-hydroxyecdysone resulted in no significant effect on transhydrogenase activity. The results of this study make evident a relationship between the presence of ecdysone and 20-hydroxyecdysone on the overall activity of M. sexta midgut mitochondrial transhydrogenations. The potential mediation of the energy-linked mitochondrial transhydrogenations involved with NADPH synthesis through the developmental relationship of ecdysone and 20-hydroxyecdysone is considered.

The molting hormones from the embryonated EGG of the tobacco hornworm, Manduca sexta (l.).

26-Hydroxyecdysone is the predominant molting hormone in 24- to 44-hour-old embryonated tobacco hornworm eggs, accounting for approximately 80% of the ecdysones present at this stage of development. This molting hormone was previously shown to be the major ecdysone present in 48- to 64-hour-old embryonated eggs of this insect. During both of these periods of embryonic development in the hornwor...

The release of the prothoracicotropic hormone in the tobacco hornworm, Manduca sexta, is controlled intrinsically by juvenile hormone.

Pupal development is elicited early in the last larval instar of the tobacco hornworm, Manduca sexta (Johannson), by a precise temporal and quantitative increase in the haemolymph titre of 20-hydroxyecdysone. This increase in the titre is referred to as the pupal commitment peak, and it occurs once the titre of juvenile hormone (JH) has dropped. If the haemolymph titre of JH remains elevated at...

Steroid-regulated Morphological Plasticity in a Set of Identified Peptidergic Neurons in the Moth Manduca Sexta

The lateral neurosecretory cells (LNCs) in the tobacco hornworm Manduca sexta undergo a switch in neurotransmitter phenotype during pupation. Concurrent with this change in function, the LNCs undergo a major morphological reorganization. This study characterizes the morphological change and its underlying cause. In the larva, the LNC has a very compact dendritic arborization in a small volume o...

cDNA cloning and expression of a hormone-regulated heat shock protein (hsc 70) from the prothoracic gland of Manduca sexta.

The brain neuropeptide prothoracicotropic hormone (PTTH) stimulates a rapid increase in ecdysteroid hormone synthesis that is accompanied by general and specific increases in protein synthesis, including that of a 70 kDa cognate heat shock protein (hsc 70). To further understand the possible roles of hsc 70, hsc 70 cDNA clones were isolated from a tobacco hornworm (Manduca sexta) prothoracic gl...

The EGPs: the eclosion hormone and cyclic GMP-regulated phosphoproteins. II. Regulation of appearance by the steroid hormone 20-hydroxyecdysone in Manduca sexta.

Two proteins (the EGPs) in the CNS of the tobacco hornworm Manduca sexta are phosphorylated by the action of the neuropeptide eclosion hormone (EH), which triggers ecdysis behavior (Morton and Truman, 1986, 1988). The onset of sensitivity to EH requires prior exposure to the steroid hormone 20-hydroxyecdysone (20-HE). A series of steroid removal and replacement experiments indicates that the EG...
